Andrea Bertotti has a scientific background in cancer biology, with a specific focus on receptor tyrosine kinase (RTK) signal transduction, oncogene addiction and targeted therapies.
As a Ph.D. student, he studied the biochemical mechanisms underlying the cooperation between RTKs and adhesion molecules during cancer progression. During his post-doc, he worked on identifying molecular traits associated with oncogene addiction, taking advantage of medium-to-large scale transcriptional and phosphoproteomic profiling approaches.
Starting 2008, he focused his research activity on more translational projects, with a particular interest in metastatic colorectal cancer. In particular, he initiated - and subsequently coordinated - a big effort aimed at generating a reliable preclinical platform for high-fidelity anticipation of clinical findings. This resulted in a collection of more than 800 liver metastases from colorectal carcinoma, which have been systematically transplanted in immunocompromised mice (patient-derived xenografts, PDXs), concomitantly profiled for molecular characteristics, and treated with approved or investigational compounds.
The informative potential of this platform, which constitutes a unique resource for translational studies in the field, led to studies that have been published in major scientific journals. A discovery with major clinical implications has been the finding that HER2 amplification predicts resistance to anti-EGFR antibodies and sensitivity to anti HER2 treatment, which recently entered the international guidelines for treatment of metastatic colorectal cancer.
More recently, thanks to an ERC Consolidator Grant funding (Grant N°724748), his research interests focused on the investigation of the molecular mechanisms sustaining drug tolerance and persistence of residual disease at maximal response to targeted therapies.
In particular, his studies focused on the identification of candidates for combination therapies with the goal of eradicating the residual disease in EGFR-dependent colorectal tumours treated with anti-EGFR blocking antibodies (i.e cetuximab). These studies resulted in a number of therapeutic approaches that have been validated in PDXs and are now considerated for the design of prospective clinical trials.
Moreover, he has been interested in deepening our understanding of the evolutionary properties of tumours, and in particolar of microsatellite stable colorectal cancer. To this aim, he is exploiting both commercial cancer cell lines and tumoroids derived from PDXs (Patient-derived tumoroids, PDTs) to study de mutation rate of colorectal tumours, both in basal conditions and under the selective pressure of targeted treatments.
